Following Kirayoshi's lead...here are my top ten moments:

10. Buffy: “World is what it is. We fight, we die. Wishing doesn't change that.”
Giles: “I have to believe in a better world.”
Buffy: “Go ahead. I have to live in this one.” - The Wish
Aren’t alternate realities fun?  Shows you just how different things could have turned out for our Scoobies.

9. Xander: “On my seventh birthday, I wanted a toy fire truck, and I didn't get it, and you were real nice about it, and then the house next door burned down, and then real fire trucks came, and for years I thought you set the fire for me. And if you did, you can tell me.” - The Replacement
Ah, the things we do for our friends. :)

8. Anya: “I don't understand! I don't understand how this all happens. How we go through this. I mean, I knew her, and then she's... there's just a body, and I don't understand why she just can't get back in it and not be dead anymore. It's stupid. It's mortal and stupid. And Xander's crying and not talking. And I was having fruit punch, and I thought, well, Joyce will never have any more fruit punch, ever. And she'll never have eggs or yawn or brush her hair. Not ever. And no one will explain to me why.” - The Body
Mentioned by others, but I think it bears repeating.  I’m not a huge Anya fan, but this made me want to reach out and hold her hand.  It’s a much different side from her usual ‘dance of capitalist superiority.’

7. Spike: “I do remember what I said. The promise. To protect her. If I'd have done that, even if I didn't make it, you wouldn't have had to jump. But I want you to know I did save you. Not when it counted, of course, but after that. Every night after that. I'd see it all again, and do something different. Faster, or more clever, you know? Dozens of times, lots of different ways. Every night, I save you.” - After Life
I really disliked neutered Spike.  In an immense way.  But this little speech kind of grabbed my heart and twisted.  To take a page out of Jet Wolf’s book, no doubt all of the Scoobies felt this way…but Spike is being Spike by saying exactly what is on his mind.  In my own little world I like to pretend it was Willow who was saying this to Buffy.

6. The whole episode of “Dopplegangland.”  ‘Nuff said.  It could make up a whole ‘Top Ten’ just by itself. 

5. Giles: “If you are Buffy, then you'll let me tie you up...without killing me...until we find out whether you're telling the truth.”
Faith (Buffy): “Giles, Faith has taken my body, and for all I know she's taken it to Mexico by now. I don't have time for bondage fun.” - Who Are You?
Wonderful episode, and this just had me in stitches.  Sarah and Eliza did such a fabulous job acting as one another’s characters. 

4. Buffy: “Does it get easier?”
Giles: “What do you want me to say?”
Buffy: “Lie to me.”
Giles: “Yes, it's terribly simple. The good guys are always stalwart and true, the bad guys are easily distinguished by their pointy horns or black hats. We always defeat them and save the day. No one ever dies, and everybody lives happily ever after.”
Buffy: “Liar.” – Lie To Me
Not everything is always black and white.  But we like to pretend it is.

3. Dawn: “I guess sometimes you can't help.”
Buffy: “So, what then? What do you do when you know that?” – Help
Season 6 and 7 were very disappointing on the whole, but this had to be one of my favorite episodes.  It reminded me of two things.  One is that destiny is often met on the road taken to avoid it.  A bit cliché, I know, but still very true.  And the second is that sometimes you can do everything right (Buffy managing to save Cassie, numerous times) and still lose.

2. Giles: "Why did you come here? This wasn't your fight."
Buffy: "Are you trying to get yourself killed? You can't leave me. I can't do this alone." - Passion
It was so heart wrenching to see Buffy holding an emotionally broken Giles.  Just as we will come to see Xander hold Willow in a similar fashion in “Grave.”  How would any of us react to someone we loved being killed?

1. Have to agree with Kirayoshi.  Buffy’s final sacrifice in ‘The Gift’ is tops for me as well.  She did what needed to be done even at the cost of her own life.  She fought the good fight and there was no more fitting way for her to go than to save those dear to her.

For the record, I thought I'd rattle off my list of my ten favorite moments of the show;
 
10) "I'm giving it away", Same Time, Same Place.  It was great to see Buffy take a giant step in rebuilding her friendship with Willow.  Oh if only Joss had followed through.  Or if SMG and AH were still on speaking terms by the end of Season Seven.  For the record, I do not blame Joss, or Marti Noxon or Jane Espenson, or any one single person or factor for the two-season-long derailment of BtVS.  I suspect there were so many mitigating circumstances involved that you couldn't count them.
 
9) "I think I'm kinda gay", Dopplegangland.  Truly the most prophetic moment in the show's history.  I especially liked how Buffy glowered Angel into silence to keep Willow from wigging.
 
8) "They made it theirs, and they had fun", Prophecy Girl.  I recommend this scene above the scene where Buffy quits because it shows how Buffy, even if she wants desperately to survive, will sacrifice everything for her friends. 
 
7) "Oxygen becoming an issue!", Dopplegangland.  Buffy and Xander went from despair to exhaltant joy in twenty seconds at the sight of Willow(I think she'd have that effect on anyone, really!) Ah, love that Buffy/Willow/Xander bonding!  We needed more of that after the show moved to UPN.  Instead we got St. Sunkencheeks!  No wonder SMG quit after Season Seven!
 
6) Buffy's return to Sunnydale, Anne.  Not the show's finest hour(although any episode would have been lacking after Becoming), but it was satisfying to see Buffy and her mom hugging again, especially after Joyce blew her cool in the second season finale. A sentimental favorite.
 
5) Spike's "Happy Meals with Legs", speech, Becoming 2.  Forget St. Sunkencheeks in Season seven, this was the quintessential Spike!  The vamp we loved to hate!  Both congenial and sadistic at the same time!  I miss him.  And I suspect James Marsters does too.
 
4) Anya's breakdown over Joyce's death, The Body.  A heartbreaking ep, and for once Anya had some character development, going beyond her "I love sex, I love money" persona.  When she begged the question 'Why?', she put the whole thing in perspective.
 
3) "It's a good fight, and I want in!", Choices.  If we believe that Buffy and Willow belong together, and they do, this is the scene that spelled it out.  Willow decides the course of her future, to stay with Buffy and fight the good fight. 
 
2) "Close your eyes", Becoming 2.  Fewer scenes show Buffy as heroically as this one, where she sent Angel to hell to stop Acaltha.  The desicion broke her heart, but in the end she did what had to be done to save the world.  In fact, only one moment in the show improved on this sublime moment...
 
1) Buffy's final sacrifice, The Gift.  The scene that got it right.  Perhaps the last moment when Buffy was truly the self-sacrificing hero she was destined to be.  She didn't throw her life away, she gave it freely to save the ones she loved.  And for the first time in her life, she felt at peace with her destiny.  For the first and only time in the show's history, Buffy was truly the Slayer.
